Methodology
Indirect immunofluorescence
Specimen Requirements
Acceptable specimens: Bronchoalveolar lavage (BAL), bronchial wash (BW), properly collected induced sputa (IS) and lung tissue.
Bronch Lavage: Physician collected specimen is a preferred sample.
Induced Sputum: Use nebulized saline to induce coughing and sputum production.
Collect specimens in sterile containers. Tissue should be submitted in ≤1 mL saline.

Additional Information
Use: Indirect immunofluorescent method for detection of P. carinii cysts and trophs in respiratory samples.
Specimen Transport Temperature and Stability
Refrigerate: 2-8° C
DO NOT add alcohol to sample.
Stability: Induced sputa are stable for 24 hours at 4°C
before testing.
BAL/BW are stable for 1 month at 4°C before testing.
Tissue: Freeze immediately or prepare touch prep and acetone fix on
slide.
